Sample Pretreatment: 1. Sample preparation
1.1 Juice drinks and pure fruit juice
Start with 10 mL sample, adjust to pH 10 - 11 with 0.1 M NaOH, add 15 mL ethyl acetate, shake 1 min, centrifuge 1 min at 4,000 rpm, collect ethyl acetate layer. Repeat 15 mL ethyl acetate extraction, and combine organic phases, vacuum distillation at 30 ºC to near dry. Dissolve residue with 0.1 M HCl (6 mL) twice.
1.2 Fruit juice concentrate
Start with 2 mL sample mixed with 8 mL H2O, adjust to pH 10 - 11 with 0.1 M NaOH, and then follow the above steps.
2. Sample purification
ProElut™ PXC 60 mg / 3 mL (Cat#68203)
Condition: 3 mL MeOH / 3 mL H2O
Load: Adding sample
Wash: 3 mL H2O / 3 mL MeOH
Elute: 3 mL MeOH (5 % NH4OH)
Reconstitute: Evaporate to near dry at 30 ºC, reconstitute to 1 mL with mobile phase
Conditions: HPLC method
Column: Inspire™ 5 µm C18, 250 x 4.6 mm (Cat#81006)
Mobile Phase: Phosphate buffer:MeCN = 75:25
Flow Rate: 1.0 mL/min
Temperature: 30 ºC
Detection: UV 288 nm
Injection Volume: 20 µL
Phosphate buffer: 1.38 g sodium dihydrogen phosphate, 1.41 g disodium hydrogen phosphate, dissolve in 1,000 mL water
