Sample Pretreatment: Samples: Single standard stock solution was prepared at 1mg/mL.When preparing a 12 compounds mixture solution, each analyte was prepared at the concentration of 25µg/mL, exceptdiphenhydramine, terfenadine and meclizine which were250µg/mL, promethazine was made at a concentration of 12.5µg/mL.
Conditions: Mobile Phase:
A: 0.1% trifluoroacetic acid in water
B:0.02% trifluoroacetic acid in 75:25(v/v) acetonitrile:methanol
Gradient: 5%B hold 0.5min, 5 to 10%B in 1.5min, 10 to 33%B in 4min, 33 to 48%B in 1.5min, 48 to 75%B in 2.5min, hold 2 min, equilibrate at 5%B for 4 min
Flow Rate: 0.3mL/min;
Injection: 10µL;
Temperature: 35℃;
Wavelength: 254nm
